FHD Mice Generate More Sequence Diversity at Comparable Titers

In a head-to-head CD22 campaign comparing Atlas FHD mice against wild-type (WT) mice, serum titers were comparable across both groups, meaning immunization conditions were equivalent. Despite identical effort, FHD mice generated significantly more IgG-producing B cells, more antigen-positive B cells, and more unique antibodies than WT mice. Of the unique CD22-positive hits recovered, 84% came from Atlas FHD mice versus 63% from WT.

More sequence diversity from the same campaign means more candidate options, without additional cost or time. Following immunization, 42 of 42 heavy chain V-genes were observed in use, not just present in the transgene, but actively expressed. Real breadth, not catalog insertion.

Epitope diversity isn’t just a discovery metric, it’s a drug development asset. Broader epitope coverage gives programs more mechanistic options and more flexibility as development advances. Using SPR-based epitope binning across all candidate antibodies recovered from the CD22 campaign, 8 distinct competition bins were identified across 17 candidates. FHD mouse sequences were represented in all 8 bins.

Strong Affinity Range With Favorable Developability Profiles

A broader sequence pool is only valuable if the resulting candidates are drug-like. Across the FHD candidate antibody set, developability profiling showed strong results across all five assessed parameters: hydrophobicity (HIC-HPLC), self-interaction (AC-SINS), polyreactivity (BVP ELISA), and thermal stability (Tm and Tagg). The majority of candidates earned green-flag scores, with very few red-flag results across any single assay.
